چکیده

Introduction Amyloid plaques are known as a marker of Alzheimer's disease. They are traditionally detected as hypointense spots on T2-weighted images taking advantage of the susceptibility effect due to iron embedded inside them. However it has been recently reported that T2 contrast might be more efficient to detect cortical amyloid plaques [1]. In this context, the goal of the present study was to compare two different strategies: (i) a conventional T2 gradient echo sequence with volume coil [2], and (ii) a zoom adiabatic T2 spin echo sequence with surface coil [1]. The images of an ex vivo APP/PS1 mouse brain acquired with both approaches were compared based on their ability to allow successful plaques detection using a home-made automatic procedure.

About the origins of enhanced visualization of amyloid plaques on APP/PS1 mouse brain using GdDOTA passive staining

Introduction T1-relaxating contrast agent (e.g. GdDOTA) “passive staining” is a smart strategy to enhance signal-to-noise ratio (SNR) for MRI microscopy (μMRI) of ex vivo tissue samples. The principle of the method is to immerse organs (e.g. brains) in a solution of GdDOTA and let it diffuse for days in order to reduce the T1 and further reduce the optimum relaxation time (TR) and accordingly t...

3D Quantitative Micro-MRI Mapping of Alzheimer’s Plaques in Transgenic Mice using Aβ1-42 Targeted-USPIOs

Introduction: Amyloid β (Aβ) plaques are one of the pathological hallmarks of Alzheimer’s disease (AD). Their visualization in the brain is very important to monitor AD progression and to evaluate the efficacy of therapeutic interventions. Numerous studies have investigated the visualization of Aβ plaques using MRI through endogenous detection both in human and in mouse brains. Only two groups ...

Detection of Amyloid Plaques Targeted by Bifunctional USPIO in Alzheimer’s Disease Transgenic Mice Using Magnetic Resonance Microimaging

Amyloid plaques are a key pathological hallmark of Alzheimer's disease (AD). The detection of amyloid plaques in the brain is important for the diagnosis of AD, as well as for following potential amyloid targeting therapeutic interventions. Our group has developed several contrast agents to detect amyloid plaques in vivo using magnetic resonance microimaging (µMRI) in AD transgenic mice, where ...

In vivo and post-mortem detection of amyloid plaques with a non-specific gadolinium contrast agent

INTRODUCTION Alzheimer’s disease (AD) is the most common type of neurodegenerative disease, but no cure has yet been found. Amyloid deposits, one of the characteristic lesions, constitute the main target for diagnostics and therapeutics and have previously been identified in mouse models using magnetic resonance (MR) microscopy. These plaques typically appear as hypointense spots on T2-weighted...

Segmentation of Amyloid Plaques in MR images of the APP Transgenic Mouse Brain using SVM

Introduction Alzheimer’s disease (AD) is linked to increased brain deposition of amyloid-beta peptides in senile plaques. Successful visualization of amyloid deposits in the APP transgenic mouse brain with MR imaging has been reported by several investigators recently. However, there are very few reports of methods for the measurement of plaque burden in the mouse brain. The number of plaques, ...
